Serum INSL3, LH, and testosterone levels were studied in different contexts. After Sustanon treatment, testosterone initially increased but then rapidly decreased due to clearance and suppressed secretion. INSL3 complemented testosterone measurements, reflecting immediate Leydig cell function reduction. Testicular suppression in transgender girls showed strong hormone suppression. In older men with prostate cancer, INSL3 decreased more than testosterone, indicating Leydig cell impairment. Orchiectomy increased LH and decreased INSL3, while Triptorelin treatment reduced all three hormones. Limited sampling and participant numbers were study limitations. Overall, serum INSL3 and testosterone were sensitive markers for testicular suppression and had potential applications in various conditions.
