After 5 nights of application, I am surprised to say that transdermal progesterone actually works for falling asleep but not for prolonging the duration of sleep. Also, it tends to decrease my daily energy/endurance, does NOT make me less depressed, does NOT make my body feel warmer, and may have a negative impact on orgasm. Read below ... I got from Amazon the Progesto-Life cream (Advanced Formula by Smoky Mountain Nutrition). The cream is a simple moisturizing base without additional herbs and contains 500mg of micronized Progesterone per ounce = 500mg/28.35g = 17.6mg progesterone per gram of cream. I applied between 1g (~ 18mg progesterone) and 0.6g (~ 10mg Progesterone) of cream on my neck (thin skin, vascular, close to brain) before bed. Without progesterone, my current HIV drug regimen makes it completely impossible to fall asleep - I just lay there napping. After I apply the progesterone cream, I start yawning and am able to fall asleep and dream. The sleep remains short though - about 6 hours, so progesterone did not correct for that. I've read online that the progesterone metabolites 5alha- and 5beta-pregnanolone bind to GABAa receptor and exert hypnotic effects. One morning I applied a second dose of the cream around 6am but it did not help me fall asleep, just made me groggy during the day. Apparently, the low transdermal dose ENHANCES the natural body cycle of falling asleep, cannot induce sleep by itself at any time of the day. I've read online that a large intravenous dose can induce sleep but we are not in that regimen at all. There were negative side effects. On the next day after the dose, I feel an obvious reduction in energy, strength, and endurance for working out and prolonged walking. There are claims online that progesterone actually suppresses adrenaline and decreases calcium uptake in smooth muscles (in blood vessels), possibly in skeletal muscles too, which would explain the energy/strength reduction. It does make me more mellow (non reactive) probably again by reduction of adrenaline. This could be good for someone in a high anxiety/stress state but definitely does not make me feel "less depressed" or "full of energy". Also, my body does not feel warmer on those doses. Sexually, I do not feel a rise in libido, erection seems the same, and I think it impairs negatively orgasm (again by reducing excitory adrenaline). In summary, progesterone at those minimal transdermal doses in males mainly helps fall sleep at night and decreases energy/endurance on the next day. This qualifies it as a mild DEPRESSANT on the central nervous system, similar to diphenhydramine or alcohol.
